چکیده

L aser capture microdissection (LCM) is a new technology that is becoming increasingly important for studies of neurodegenerative disorders. A characteristic feature of all neurodegenerative diseases is “selective vulnerability.” In each of the disorders, there is selective degeneration of particular types of neurons, with relative preservation of much of the rest of the brain. Familiar examples are the selective degeneration of dopaminergic neurons in Parkinson disease, the striking depletion of basal forebrain cholinergic neurons in Alzheimer disease, the selective atrophy of the caudate nucleus in Huntington disease, and the loss of spinal motor neurons in amyotrophic lateral sclerosis. Closer examination reveals an even more finegrained pattern of neuronal injury. For example, in Parkinson disease there is striking depletion of dopaminergic neurons in the ventral tier of the substantia nigra pars compacta, with very little injury to dopaminergic neurons located only a few millimeters away in the dorsal tier of the nucleus. In Huntington disease, there is an exquisite degree of selectivity among the different types of neurons found within the caudate and putamen, with striking depletion of the medium spiny projection neurons, especially those projecting to the globus pallidus, in contrast to preservation of the intermingled interneurons. These characteristic patterns of injury are evident even in genetically determined forms of neurodegenerative disease where the fundamental defect can be shown to be present in all neurons. Understanding the basis for these patterns of neuronal loss may provide important insight into the mechanisms of neurodegeneration.

A novel ex vivo Huntington’s disease model for studying GABAergic neurons and cell grafts by laser microdissection

Organotypic brain slice cultures have been recently used to study neurodegenerative disorders such as Parkinson's disease and Huntington's disease (HD). They preserve brain three-dimensional architecture, synaptic connectivity and brain cells microenvironment. Here, we developed an innovative model of Huntington's disease from coronal rat brain slices, that include all the areas involved in the...

Production of RNA for Transcriptomic Analysis from Mouse Spinal Cord Motor Neuron Cell Bodies by Laser Capture Microdissection

Preparation of high-quality RNA from cells of interest is critical to precise and meaningful analysis of transcriptional differences among cell types or between the same cell type in health and disease or following pharmacologic treatments. In the spinal cord, such preparation from motor neurons, the target of interest in many neurologic and neurodegenerative diseases, is complicated by the fac...

Laser capture microdissection in the genomic and proteomic era: targeting the genetic basis of cancer.

The advent of new technologies has enabled deeper insight into processes at subcellular levels, which will ultimately improve diagnostic procedures and patient outcome. Thanks to cell enrichment methods, it is now possible to study cells in their native environment. This has greatly contributed to a rapid growth in several areas, such as gene expression analysis, proteomics, and metabolonomics....

Laser capture microdissection: methodical aspects and applications with emphasis on immuno-laser capture microdissection.

Laser capture microdissection (LCM) is an easy, extremely fast and versatile method for the isolation of morphologically defined cell populations from complex primary tissues for molecular analyses. However, the optical resolution is limited due to the use of dried sections without coverslip necessary for tissue capture, and routine stains such as hematoxylin and eosin are sometimes insufficien...
